Sulfate-reducing bacteria (SRB) found in oilfield water systems can be a serious threat to oil productivity and profits. If left unmonitored and undetected, these microorganisms create hazardous hydrogen sulfide and can cause serious damage to infrastructure through microbiologically influenced corrosion (MIC). Bacterial corrosion can cause structural failures and foul or plug pipes, pumps and other equipment. SRBs can also sour an oil reservoir, requiring expensive remediation.
Conventional SRB testing methods can take weeks to deliver results – giving SRBs more time to multiply and cause damage such as biocorrosion. The 3M™ Rapid SRB Detection Pouch is a standalone culturing method designed to make field testing easy, accurate and – best of all – fast. Testing has shown our detection pouch can provide results in significantly less time than conventional methods like serial dilution bottles. Light and compact, the bacteria test kit is so small, 200 tests fit easily in a backpack for portability in the field or offshore.

Test Process

What should I use to inoculate the sulfate reducing bacteria test?
The best way to inoculate is with a 3mL sterile, individually wrapped, disposable transfer pipette. These are available from any lab supply company.

Are serial dilutions required for the pouch?
No. One pouch is used per SRB test and provides a quantitative result.

Does the pouch need to be incubated?
Incubate the test at 30°C. If the sample temperature is recorded at higher than 30°C, incubate within 5°C of the recorded sample temperature per NACE TM 0194 -2014. Incubating below ideal temperature may cause slower, underestimated, or false negative test results.

How do I dispose of inoculated pouches?
Follow current local, national and industry standards for disposal.

What is the shelf life and storage requirements of the Rapid Sulfate Reducing Bacteria (SRB) Detection Pouch?
For maximum shelf life, store unopened foil packages in a refrigerator (2-8C). Pouches can be stored for 12 months in room temperature conditions (25C), it will maintain its function after short term exposures of up to 1 month at to conditions 45°C (113°F).

Can the pouch provide quantitative results?
Yes! The pouch uses time-based quantitation to provide fast, accurate results. Using the interpretation guide, a user can approximate the concentration of SRBs based on the time it takes the test to turn black after inoculation, typically in two days or less for samples with >10 SRB/mL.

Are the 3M™ Rapid SRB Detection Pouch results compatible with my old bug bottle results?
Yes. 3M™ Rapid SRB Detection Pouch results will not match bug bottle results exactly but can be reported in the same units of SRB CFU/mL of sample.

How can I ensure the results are objective?
Each box of pouches comes with an interpretation card that uses a calibrated grayscale key to provide objectivity between operators.

Does the pouch work at elevated temperatures?
Yes. SRBs have been cultured in the pouch at incubation temperatures of up to 80°C (176°F).

Does the pouch work with samples containing high Total Dissolved Solids (TDS)?
Yes. The pouch is designed for samples of any TDS level. Unlike serial dilution bottles where matching salinity is important, the 3M™ Rapid SRB Detection Pouch can be used for samples of any salinity.

What interferes with the pouch?
The 3M™ Rapid SRB Detection Pouch has been validated against many common oilfield chemicals. See more in this NACE paper(link). The only chemical likely to interfere is hydrogen sulfide, which will turn the test black in less than 5 minutes at >25 ppm.

What measures should be taken for high hydrogen sulfide samples?
Dilute the sample before inoculation if H2S is present at >25 ppm. Directions can be found in the product user guide and how-to video.

Which SRB species does the pouch detect?
Lab validations were completed using Desulfovibrio desulfuricans and Desulfovibrio vulgaris. Additionally, field and lab validations included diverse SRB species such as thiosulfate reducers and thermophilic SRBs.
